청미래 덩굴 초임계 추출물의 멜라닌 생성억제 효과 (Inhibition of Melanogenesis by Smilax china L. Superchritical Extract)

초록

The purpose of this study is to investigate the effect of supercritical extract of Smilax china L. on the β-catenin activity involving Wnt signaling and melanogenesis inhibition. The yield coefficient of supercritical extract from the leaves (SCL) was four times higher than that from the roots (SCR). SCL of 100 μg/ml had almost no cytotoxicity, while SCR showed a low cytotoxicity. SCL showed a melanogenesis inhibitory effect in a dose-dependent manner for α-MSH and Wnt3a treated melanoma cells, within the concentration of 100 μg/ml. SCR of 20 μg/ml concentration showed the highest inhibitory effect of melanogenesis. Both SCL and SCR inhibited the β-catenin activity of SW480(human colon cancer cell) in a dose-dependent manner. In the SCL treated B16F10 mouse melanoma cells, the expression of MITF and Tyrosinase protein was decreased in a concentration-dependent manner. SCL seems to inhibit the melanogenesis in B16F10 cells by inhibiting the expression of MITF through the α-MSH)/ melanocortin 1 receptor (MC1R) signaling pathway. From these results, SCL may be used as a raw material in the production of skin-whitening cosmetics.
